Based in North Carolina, US, and founded in 1998, Tranzyme Pharma, Inc. focused on creating advanced drugs for digestive disorders with high unmet medical needs that targeted specific hormones involved in digestion to address issues like delayed stomach emptying and diarrhea. In February 2012, Tranzyme Pharma received an additional USD 9. 3 million in term loan proceeds through an amendment and restatement of its existing debt financing facility with Oxford Finance LLC and Horizon Technology Finance Corporation.

As of October 2000, the company was led by CEO Vipin K. Garg. Tranzyme Pharma's products include ulimorelin, an intravenous prokinetic agent designed to address postoperative ileus and acute gastric motility disorders, and TZP-102, an oral prokinetic agent for treating gastroparesis. Both products are potent ghrelin agonists and are in late-stage development.

Additionally, Tranzyme is advancing TZP-201, a motilin antagonist aimed at treating moderate-to-severe diarrhoea, and TZP-301, a ghrelin antagonist for metabolic diseases. Tranzyme MATCH technology facilitates the construction of synthetic libraries of drug-like, orally administered macrocyclic compounds predictably and efficiently. The company also offers HitCreate, a library of highly diversified small-molecule macrocycles created using MATCH technology. HitCreate contains novel, drug-like compounds with distinct topologies and diverse functionalities, including lipophilic, polar, and charged groups.
